The Glasgow Warrior's opening try proved the difference in an uninspiring first half and he crossed again in the 55th minute. Greig Laidlaw had gone over the line after the first of Kenki Fukuoka's two tries for Japan to take him past the 200-point mark in his 22nd Test, while Alasdair Dickinson, Duncan Weir and Sean Lamont all crossed when the visitors were down to 14 men.
However, it was Seymour's performance which caught the eye and the 25-year-old is hoping Scotland can now go on to bigger and better things.
"In the first half to get two tries is really special for me," he told the BBC. "My parents were in the stand.
"We're not happy with a couple of things but I'm not going to complain about today. To get two tries is the icing on the cake. We're building for the World Cup in 2015 and I would like to think we can have hope for the future."
Scrum-half Laidlaw was also delighted with the result, but feels there are still things which the squad have to work.
He said: "It's a great win for us. We're all annoyed at giving away soft tries but we scored some nice tries and we'll take the result for our first game.
"You can only beat the players in front of you, but we'll have a look at the tries conceded."
Lock Tim Swinson was another debutant at Murrayfield and paid tribute to Japan for refusing to go down easily.
He said: "It wasn't going to be an easy day. The Japanese are a quality Test side, they showed that today."
